Abstract SCOPE: Insulin resistance represents an independent risk factor for metabolic and cardiovascular diseases. Researchers have been interested in identifying active harmless compounds, as many insulin-sensitizing drugs have shown unwanted side-effects. It has been demonstrated that anthocyanins and one of their representative metabolites, protocatechuic acid (PCA), ameliorate hyperglycemia, and insulin sensitivity. This study investigated the mechanism of action of PCA responsible for the glucose uptake upregulation. METHODS AND RESULTS: In human visceral adipocytes, PCA stimulated insulin receptor substrate-1 (IRS-1) tyrosine phosphorylation (+40% with respect to untreated cells) and the downstream events, i.e. phosphoinositide 3-kinase binding to IRS-1 and Akt phosphorylation (+100%, +180%, respectively, with respect to untreated cells). The insulin-like activity of PCA seemed to be mediated by insulin receptor since by inhibiting its autophosphorylation, the PCA effects were completely abolished. Furthermore, PCA was able to activate adenosine monophosphate-activated protein kinase, a serine/threonine kinase whose activation elicits insulin-sensitizing effects. CONCLUSION: This study showed that PCA stimulates the insulin signaling pathway in human adipocytes increasing GLUT4 translocation and glucose uptake. Decreasing insulin resistance is a most desirable aim to be reached for an effective therapeutic/preventive action against metabolic syndrome and type 2 diabetes. Cinnamon is a spice from the dried aromatic bark of the cinnamon tree. It is used as a spice for all sorts of cookery and is lately added to some bodybuilding supplements. Creatine monohydrate has without a doubt become one of the most popular nutritional supplements, and it is used to enhance athletic performance. Companies strive to develop formulations in order to maximize creatine absorption and may also provide a means to either partially bypass or up-regulate the function of creatine transporter-1. It has been observed that cinnamon extract (Cinnulin) mimics the effects of insulin, thereby up-regulating glucose uptake and insulin signaling. [1] It has been observed in the literature that Cinnamon extract (Cinnulin™) potentiates an insulin effect and successive insulin signaling through the up-regulation of glucose transport.[2] So, combining creatine monohydrate with Cinnamon extract (Cinnulin™) may increase muscle creatine uptake by stimulating skeletal muscle-mediated glucose uptake. A study in rats suggested that the cinnamon extract would improve insulin action via increasing glucose uptake, at least in part through enhancing the insulin-signaling pathway in skeletal muscle.[3] Cinnulin appears to act like insulin, which may up-regulate insulin signaling. A research done by Ryan D. Schoch [1] examined the effects of seven-week Cinnulin and Creatine monohydrate supplementation. Muscle strength and mass, body composition, muscle creatine uptake, serum insulin and IGF-1 were measured.
